摘要:
利用改良的高效液相色谱法(HPLC)对磷酸替米考星可溶性粉的代谢物替米考星在肉鸡组织中的残留情况进行了检测分析。试验结果显示:流动相乙腈-四氢呋喃-二丁胺磷酸盐缓冲液-水的体积比为10:5:5:80。该方法对替米考星的平均回收率为77.02%~115.04%。肌肉、肝脏和肾脏中替米考星的检测限分别是0.010、0.025和0.025 μg·g-1,定量限分别是0.020、0.050和0.050 μg·g-1。以16 mg·kg-1剂量连续口服给药7 d磷酸替米考星可溶性粉,在停药后4 h到停药后1 d,替米考星在肉鸡体内肝脏、肾脏和肌肉组织中的残留即出现显著下降;其中,在肝脏中的残留最高,其次是肾脏,而肌肉组织内的残留较低。经休药期计算可知,替米考星在肝脏、肾脏和肌肉组织的休药期分别为9.6、12.8和13.96 d,按双单侧95%置信区间计算替米考星在肉鸡的休药期应该为14 d。
Abstract:
The elimination regularity and residues of tilmicosin phosphate in broiler tissues were detected at UV 280 nm by using improved high performance liquid chromatography(HPLC),with the proportion of acetonitrile,tetrahydrofuran,dibutylamine phosphate buffer and water was 10:5:5:80 by the mobile phase.The results showed that the average recovery rates of tilmicosin were 77.02%-115.04% and the detection limit of tilmicosin in muscle,liver and kidney were 0.010,0.025 and 0.025 μg·g-1,respectively.The quantitation limits in muscle,liver and kidney were 0.020,0.050 and 0.050 μg·g-1,respectively.After 7 days of oral administration at dosage of 16 mg·kg-1,the tilmicosin residues in liver,kidney and muscle tissues of broiler decreased significantly within 4 hours to 1 day after discontinuation.The highest level of tilmicosin residues occurred in liver and followed by kidney and the lowest in muscle tissues.According to the calculation by software WT 1.4,the withdrawal period of tilmicosin was 9.6,12.8 and 13.96 days in liver,kidney and muscle respectively,suggesting that the withdrawal period of tilmicosin phosphate for broiler using should be 14 days by the 95% confidence interval analysis.
