摘要:
[目的]为仔猪生产筛选适宜添加的油脂和剂量。[方法]将20窝初生日龄和初始体质量相近的7日龄长大二元杂交仔猪,随机分为基础日粮(CK)和4个处理组(T1~T4),每组4窝,每窝不少于8头。试验分为哺乳期(7~21 d)和断奶期(22~35 d)2个阶段,其中CK组均饲喂基础日粮,T1组在基础日粮上分别添加质量分数为0.16%和4%乳化椰子粉,T2组在基础日粮上分别添加0.32%和8%乳化椰子粉,T3组在基础日粮上分别添加0.16%乳化椰子粉和4%豆油,T4组饲喂基础日粮和在其基础上添加4%豆油。试验过程中,测定仔猪生长性能和血清脂质指标,并检测肝脏乙酰辅酶A乙酰基转移酶(ACAT)、3-羟-3-甲基戊二酸单酰辅酶A还原酶(HMGCR)和胆固醇7α-羟化酶(CYP7A1)的活性变化,分析HMGCR、CYP7A1和低密度脂蛋白受体(LDLR)基因(mRNA)相对表达量变化。[结果]与对照相比,各处理对2个阶段仔猪生长性能均无显著影响(P>0.05),添加不同剂量乳化椰子粉对哺乳阶段仔猪血清脂质指标无显著影响(P>0.05),T2组能显著提高断奶阶段仔猪血清总胆固醇含量(P<0.05)。同时,T2较T1组断奶仔猪血清总胆固醇含量显著提高(P<0.05)。与对照相比,各处理HMGCR、CYP7A1和ACAT酶活性和CYP7A1、LDLR、HMGCR基因表达量无显著改变(P>0.05),但T2组有增加HMGCR酶活性和HMGCR基因表达量趋势(P<0.1)。[结论]添加不同剂量和类型油脂对试验仔猪生长性能无显著影响,但高剂量的乳化椰子粉能显著提高血清总胆固醇含量,并有增加肝脏合成胆固醇关键酶HMGCR酶活性和HMGCR基因表达的趋势。
Abstract:
[Objectives]This experiment was conducted to select a suitable amount and type of oil for piglets. [Methods]In this experiment,12 litters of 7 days of age healthy piglets with similar body weights and birthdates were selected and randomly divided into five test groups which were labeled as CK,T1,T2,T3 and T4,respectively. We treat CK as the control group and the rest is treatment group. Each group was 4 nests and 8 piglets in each nest at least. Piglets were weaned at 21 days of age,the test was divided into two periods,piglet’s age of the first period was 7-21 days,and that of the second period was 22-35 days. In the first and second period,CK just provided basal feed. T1 provided 0.16% and 4% emulsified coconut powder to the basal feed,respectively. T2 provided 0.32% and 8% emulsified coconut powder to the basal feed,respectively. T3 provided 0.16% emulsified coconut powder and 4% soybean oil to the basal feed,respectively. T4 provided basal feed and 4% soybean oil to the basal feed,respectively. Growth performance and serum lipid index were measured. Liver cholesterol metabolism related enzyme activities of acetyl coenzyme A acetyltransferase(ACAT),3-hydroxy-3-methylglutaryl coenzyme A reductase(HMGCR)and cholesterol 7 alpha-hydroxylase(CYP7A1)of piglets were analyzed. The expression of mRNA of HMGCR,CYP7A1 and low density lipoprotein-receptor(LDLR)in liver were determined by real time PCR. [Results]Compared with CK,growth performance have no statistical difference(P>0.05)in treatment groups during the first and second period. By adding different doses of emulsified coconut powder,there is no difference between treatment group and control group in piglet serum lipid index during the first period. Compared with CK,8% emulsified coconut powder has significantly improved the piglets serum total cholesterol(P<0.05)during the second period. Meanwhile,compared with T1,8% emulsified coconut powder has significantly improved the piglets serum total cholesterol during the second period. During the second period,compared with CK,liver cholesterol metabolism related enzymes activities of ACAT,HMGCR and CYP7A1,the expression of mRNA of CYP7A1,LDLR and HMGCR have no statistical difference in treatment groups(P>0.05),while enzymes activity and mRNA expression level of HMGCR in 8% emulsified coconut powder group has an trend(P<0.1)to be improved. [Conclusions]Different dosage and source dietary fats have no different effect on the growth performance of piglets. High dosage medium-chain fatty acids can increase the serum levels of total cholesterol and has an trend to improve HMGCR enzymes activity and HMGCR mRNA expression level.
