摘要:
本文采用高效液相色谱(HPLC)与气相色谱(GC)和生物分析相结合的方法纯化葡萄中β-damascenone前体。葡萄提取物用乙腈-水(82:18V/V)作流动相,在Lichrosorb NH_2柱上色谱分离,并分部收集柱洗脱物。然后,用气相色谱和生物分析法测定洗脱物水解产物中的β-damascenone,确定它前体的高效液相色谱保留时间。在相同的高效液相色谱条件下,收集前体色谱峰相应的柱洗脱物。经过2~3次纯化后,可得到3.2mg纯度相当高的β-damascenone前体。
