摘要:
采用高效液相色谱-二极管阵列检测器(HPLC-DAD)法,测定了云南省18个县(区)4个主栽烤烟品种(云烟85、云烟87、K326和红花大金元)的多酚物质,建立了云南烤烟多酚物质的HPLC指纹图谱。指纹图谱检测的色谱条件是:Beckman Coulter USPHODS(4.6 mm×250 mm,5μm)色谱柱,流动相为甲醇和0.05 mol.L-1KH2PO4混合液,梯度洗脱,检测波长345 nm,流速0.5 mL.min-1,柱温为室温。应用相似度、相对误差等参数对所得指纹图谱进行了评价。结果表明:所建立的烤烟多酚物质指纹图谱检测方法操作简单,重复性好,响应值高,分离度好;不同产地来源、不同品种和不同叶位的指纹图谱相似度极高,但峰面积存在明显差异。因此所构建的指纹图谱可在对云南烤烟产地、品种和叶位的识别及质量控制方面发挥一定的作用。
Abstract:
To assess the possibility of the HPLC fingerprint of polyphenols in flue-cured tobacco in Yunnan, China, a useful method to control and evaluate tobacco quality was established. The polyphenols of flue-cured tobacco samples collected from 18 different places in Yunnan Province were determined by HPLC-DAD. Beckman Coulter USPH ODS column(4.6 mm×250 mm, 5 μm)was selected to seperate the polyphenols in a gradient elution mode and the mixtures of methanol and 0.05 mol·L-1 KH2PO4 was mobile phase. The flow rate was set at 0.5 mL·min-1.The column temperature was set at 25 ℃ and the detecting wavelength was 345 nm. The similarity coefficients and RSD(relative standard deviation)were used to evaluate the fingerprint method. The results showed that a simple, high response and specific method with good resolution and repeatability was well established. There were different polyphenol contents in samples from different sampling places, cultivars and leaf position, but there were high similarity produced in individual area. In conclusion, the established fingerprint method could be used for the quality control and to identify sampling place, culitivars and leaf position of flue-cured tobacco in Yunnan.
