摘要:
以不结球白菜品种苏州青(耐热)和矮脚黄(不耐热)幼苗为材料进行热激处理(40℃,0、6、12、24、48、72和96 h),测定了热激处理对不结球白菜叶片中保护酶活性、丙二醛(MDA)含量以及膜透性的影响,同时利用实时定量PCR(qPCR)技术检测叶片中BcHsp1和BcHsp2耐热基因的表达变化。结果表明:热激处理后,耐热品种叶片中SOD、POD和CAT活性的增幅大于不耐热品种;不耐热品种叶片中丙二醛含量和电解质渗透率的增幅大于耐热品种;相对于不耐热品种,耐热品种热激蛋白基因具有明显稳定的热激诱导表达特性。
Abstract:
The heat shock treament(40℃, 0, 6, 12, 24, 48, 72 and 96 h) was conducted on two non-heading Chinese cabbage varieties, Suzhouqing and Aijiaohuang, which had different heat resistant ability. The activity of leaf protective enzymes, MDA content and membrane permeability were tested in seedling stage. Real-time quantitative PCR(qPCR) was used to detect the expression of BcHsp1 and BcHsp2.The results showed that, after heat shock treament, the activity of protective enzymes in the leaves of heat resistant variety had a higher increase than those in the heat sensitive variety, and the increases of MDA content and electrolyte leakage rate were higher in the heat sensitive variety than those in the heat resistant variety. And the heat-tolerant genes were more stably expressed in heat resistant varitey than those in heat sensitive variety.
