摘要:
以142头公牛为研究对象,以HSP为耐热性状的候选基因,分别扩增125和233 bp 2个片段,采用PCR-SSCP方法检测HSP基因多态性,并对PCR产物测序和序列分析。结果表明:HSPA8存在T3358C转换突变,该突变使缬氨酸(Val)变为异亮氨酸(Ile),产生2种基因型AA和AB,频率分别为0.577 5和0.422 5;HSPA1A发现A6201C和T6202A颠换突变,其中T6202A突变使苯丙氨酸(Phe)变为异亮氨酸(Ile),产生了5种基因型AA、AB、BB、AC和BC,频率分别为0.345 1、0.493 0、0.084 5、0.035 2和0.042 2。应用SPSS17.0软件采用最小二乘拟合线性模型,分析基因座位不同标记基因型与后裔测定性能和精液品质的相关性,结果表明:1)在HSPA8,AA基因型个体产量育种值显著高于AB型(P<0.05);在HSPA1A,BB基因型个体产量育种值显著高于AC型(P<0.05),AB基因型个体蛋白育种值显著高于AC型(P<0.05)。2)在HSPA8,精子密度、精子活力和顶体完整率AA均显著高于AB(P<0.05),而精子畸形率AA显著低于AB(P<0.05);在HSPA1A,精子密度BC极显著低于AA(P<0.01),精子活力AC极显著低于AB(P<0.01),而精子畸形率AB极显著高于AC(P<0.01)。
Abstract:
In this experiment,125 bp and 233 bp fragments of heat shock protein gene(HSP)named HSPA8 and HSPA1A from 142 bulls were amplified by polymerase chain reaction(PCR),respectively.The genetic polymorphisms in the population were detected by single-strand conformational polymorphism(SSCP),and the PCR products were sequenced and sequence analysis was performed.The results indicated that T3358C transition mutations in HSPA8,and A6201C and T6202A transvertion mutations in HSPA1A were founded.T3358C mutation resulted in an amino acid change of valine into isoleucine in HSPA8,and two genoytypes (AA and AB)were found with frequency was 0.577 5 and 0.422 5,respectively.T6202A mutation resulted in an amino acid change of phenylalanine into isoleucine at HSPA1A,and five genotypes(AA,AB,BB,AC,BC)were found with frequency was 0.345 1,0.493 0,0.084 5,0.035 2 and 0.042 2,respectively.The association between different marker genotypes and progeny performance and semen quality was evaluated by least squares linear model using SPSS17.0 software.The results showed that:1)The estimated breeding value of milk(EBVM)with genotype AA was significantly higher than that with genotype AB(P<0.05)in HSPA8. Meanwhile,the EBVM of individuals with genotype BB was significantly higher than that with genotype AC in HSPA1A(P<0.05); the estimated breeding value of protein(EBVP)of individuals with genotype AB was significantly higher than that with genotype AC(P<0.05).2)Sperm density(SD),sperm motility(SM)and acrosomal integrity(AI)of individuals with genotype AA were significantly higher than those with genotype AB(P<0.05)in HSPA8,and the sperm abnormality rate(SAR)of individuals with genotype AA was significantly lower than that with genotype AB(P<0.05),however,the SD of individuals with genotype BC was significantly lower than that with genotype AA(P<0.01)in HSPA1A,SM of individuals with genotype AC was significantly lower than that with genotype AB(P<0.01),and SAR of individuals with genotype AB was significantly higher than that with genotype AC(P<0.01).
