摘要:
[目的]脱氧雪腐镰刀菌烯醇(DON)是饲料及原料中广泛存在的真菌毒素,本文以幽门结扎大鼠为模型研究DON对大鼠胃液组成的影响。[方法]将24只禁食Wistar大鼠随机分为4组,分别以0、1、5和25 mg·kg-1的DON灌胃染毒0.5 h后进行幽门结扎术,4 h后收集胃液。测定胃液体积、pH值、胃蛋白酶含量及活性、黏蛋白和金属离子含量等。[结果]与对照组相比,1、5和25 mg·kg-1 DON染毒大鼠胃液体积分别下降了24.88%、51.39%和60.88%(P<0.05),pH值升高到3.20、3.81和6.70(对照组为1.93),胃液中黏蛋白含量分别下降了8.42%、10.02%和12.30%,胃蛋白酶含量分别下降了7.96%、18.33%和50.83%,胃蛋白酶活性分别降低了27.97%、55.36%和67.82%。胃液内容物干质量显著升高,1、5和25 mg·kg-1 DON染毒组分别比对照组增加了42.97%、250.16%和2 460.32%。感应耦合等离子体发射光谱法(ICP-AES)测定发现DON染毒对大鼠胃液K+、Na+、Ca2+和Zn2+等常见金属离子含量造成不同程度影响。[结论]DON染毒明显改变了Wistar大鼠胃液组成,造成了大鼠胃分泌功能的紊乱。
Abstract:
[Objectives] Deoxynivalenol(DON) is a common mycotoxin in feed and its row materials in the world. This study aimed to explore the influence of DON on the composition of gastric juice in rats. [Methods] 24 Wistar rats were gavaged at the dose of 0,1,5 and 25 mg·kg-1 DON after fasting for 24 h,and 0.5 h later pylori ligation was used to collect gastric juice within 4 h. The composition of gastric juice was detected,including volume,pH value,pepsin content and activities,mucin and metal ions. [Results] The results showed that the DON exposure resulted in a decrease by 24.88%,51.39% and 60.88%,respectively,compared to the control,and the differences were significant(P<0.05). In addition,pH value reached 3.20,3.81 and 6.70 in DON exposed rats,with control group at 1.93. Similarly,the mucin decreased by 8.42%,10.02% and 12.30%,respectively. The contents of pepsin decreased by 7.96%,18.33% and 50.83%,and the activities decreased by 27.97%,55.36% and 67.82% at 1,5 and 25 mg·kg-1 DON group,respectively. The dry weight was significantly higher than the control,with 42.97%,250.16% and 2 460.32% at 1,5 and 25 mg·kg-1 DON,respectively. The results of inductively coupled plasma atomic emission spectrometry(ICP-AES) assay showed that DON exposure also disrupted the composition of K+,Na+,Ca2+,Zn2+ of gastric juice. [Conclusions] These results suggest that DON exposure influences the composition of gastric juice in Wistar rats,and causes the disorder of gastric secretion in rats.
