摘要:
[目的]从江苏如皋水稻田土样中分离到昆虫线虫一新株系N3(简称虫株N3),通过形态学和分子生物学特征对其进行了鉴定。[方法]利用光学显微镜和扫描电镜观察线虫的形态特征,对其形态特征进行测计并构建基于18S rDNA序列的系统发育树等,对虫株N3进行了系统分类鉴定。[结果]虫株N3主要形态特征是:食道基球退化;交合刺整体弧形弯曲,基部愈合弯曲;引带近腹部端,附着于交合刺基部;生殖乳突P3近泄殖腔,P2、P4远泄殖腔;双子宫。18S rDNA分子系统树表明:虫株N3与Diplogasteroides nasuensis最为相近,具有93.2%的同源性。虫株N3可以根据食道基球与交合刺基部的形态和引带附着位置加以区分。[结论]结合形态学和分子生物学特征鉴定,虫株N3为新种,命名为如皋似双胃线虫(Diplogasteroides rugaoensis n.sp.)。
Abstract:
[Objectives]An insectivorous nematode population N3(Hereinafter referred to as:N3)was collected from paddy field of Rugao,Jiangsu Province. Morphological and molecular biological methods were used for identification of the nematode population. [Methods]N3 was observed by scanning electron microscopy and light microscopy examination. Morphological characteristics of N3 were measured and calculated. Taxonomic status of N3 was analyzed by building phylogenetic trees based on 18S rDNA sequences. [Results]The morphological characters of N3 are mainly as follows:terminal bulb is degenerated. Spicules is overall arc bending and spicules base is coalescent;near the belly side of gubernaculum the base of spicule is attached;the third pairs of genital papillae is close to the anus,and the second and fourth pairs of genital papillae are far away from the anus;double uterus. Phylogenetic tree of 18S rDNA indicated that N3 belongs to the genus Diplogasteroides and has the most close relationship with D.nasuensis. The 18S rDNA sequences of the two nematodes share 93.2% identity. N3 can be distinguished on the basis of the different form of terminal bulb and the base form of spicule,adherent position of gubernaculum. [Conclusions]Morphological characteristics combined with molecular data showed that this nematode is a novel species of the genus Diplogasteroides,named as Diplogasteroides rugaoensis n.sp..
