摘要:
[目的]本文旨在研究60Co-γ辐照对猪血清白蛋白(PSA)免疫原性和蛋白构象的影响。[方法]应用表面等离子共振(SPR)技术、圆二色(CD)光谱、紫外光谱和荧光光谱方法,研究在(10±0.5)℃条件下,1、3、5、7和10 kGy辐照剂量处理对过敏原猪血清白蛋白免疫原性及蛋白构象的影响,并分析过敏原免疫原性与其蛋白质构象的相关性。[结果]过敏原PSA在辐照剂量低于3 kGy时其免疫原性略有降低,之后随着辐照剂量的增加而显著降低(P<0.05)。辐照对过敏原PSA构象的影响则采用CD光谱、紫外光谱和荧光光谱来进行表征。过敏原PSA经辐照处理后其α-螺旋含量明显降低,二级结构发生改变;过敏原猪血清白蛋白的紫外吸收值随着辐照剂量的增加呈上升趋势;而过敏原猪血清白蛋白的荧光光谱强度随着辐照剂量的增加而降低,其最大发射峰还发生蓝移现象。[结论]60Co-γ辐照处理可以通过改变过敏原猪血清白蛋白的蛋白质构象来达到降低其抗原性的目的。
Abstract:
[Objectives] The effect of 60Co-γ irradiation treatment on the immunogenicity and conformation of the porcine serum albumin(PSA)was investigated. [Methods] A study was carried out about the gamma irradiation treatment(1, 3, 5, 7 and 10 kGy)at(10±0.5)℃ on the immunogenicity and conformation of PSA using surface plasmon resonance(SPR), circular dichroism(CD)spectroscopy, UV spectroscopy, and fluorescence spectroscopy. The correlation between the immunogenicity and protein conformation was analyzed. [Results] The immunogenicity of PSA was slightly reduced at doses lower than 3 kGy, and then began to significantly decrease as the irradiation dose increased(P<0.05). The conformational changes in irradiated PSA were measured by using CD, UV and fluorescence spectroscopy, respectively. Gamma irradiation treatment led to a reduction in α-helices of PSA secondary structure;UV absorbance of PSA was elevated as the irradiation dose increased;the relative fluorescence intensity peak of PSA was lowered with the increase of the irradiation dose, and a blue shift of maximum emission wavelength was also observed after gamma irradiation. [Conclusions] 60Co-γ irradiation treatment could reduce the antigenicity of allergen PSA through changing their spatial conformation.
