摘要:
[目的]本试验旨在分离、多向诱导分化并鉴定犬脂肪间充质干细胞。[方法]无菌条件下采取经过健康筛查的犬腹部脂肪,机械剪碎,采用1 g·L-1胶原酶Ⅰ型消化,用DMEM+10% FBS培养,观察细胞形态特征,流式细胞术分析细胞表面抗原标志的表达,并检测其体外成脂、成骨、成软骨分化能力。[结果]运用酶消化法可以从犬新鲜脂肪组织中分离到脂肪间充质干细胞,流式分析结果显示其高表达CD29(99.9%),低表达CD34(0.5%)和CD45(0.3%)。这些细胞在体外经诱导可以分化成为骨、软骨和脂肪。[结论]犬腹部脂肪组织中分离得到脂肪间充质干细胞,这些细胞具有多向分化能力。
Abstract:
[Objectives]This study was aimed to isolate,characterize,and differentiate canine adipose-derived mesenchymal stem cells(ADSC).[Methods]Subcutaneous fat tissues from canine abdomens were obtained via germ-free procedures,mechanically shredded and digested with 1 g·L-1 collagenaseⅠ. Subsequently,ADSC were cultured in DMEM medium containing 10% FBS. Then,cell morphology was observed under microscope and cell surface antigen expression was analyzed by flow cytometry,respectively. Finally,osteogenic,adipogenic and chondrogenic differentiations were determined.[Results]ADSC was successfully isolated from fresh fat tissues of canine using enzyme digestion method. High expression of CD29(99.9%)and low expression of CD34(0.5%),CD45(0.3%)were observed in this cell by flow cytometry analysis. The ADSC could be differentiated into adipocytes,osteoblasts and chondrocytes in vitro.[Conclusions]The results indicate that ADSC can be isolated from the subcutaneous fat tissue of canine and have multi-directional differentiation ability.
