摘要:
[目的] 本文旨在研究影响无花粉污染百合杂交亲和性的相关因素,为选育无花粉污染百合新品种奠定基础。[方法] 选取无花粉亚洲百合品种‘小小的吻’和‘平凡生活’,不散粉品种‘简单华尔兹’以及花粉团聚状OT百合品种‘耶罗琳’作为母本,以11份百合种质资源为父本,进行常规杂交,研究各个组合的杂交亲和性。通过花粉管荧光显微观察与石蜡切片重点研究了‘小小的吻’为母本的杂交组合杂交亲和性差异的原因。[结果] 父本中,东方百合‘索邦’的花粉萌发率最高(95.30%),LA百合‘莫泽尔’的花粉萌发率最低(6.79%)。33个杂交组合中,以‘小小的吻’作母本的10个杂交组合的平均杂交亲和指数最高,为3.61%,其中‘小小的吻’ב小飞碟’杂交亲和指数最高;以‘平凡的生活’为母本的9个杂交组合的平均杂交亲和指数为0.90%,其中‘平凡生活’ב正直’的杂交亲和指数最高;以‘简单华尔兹’为母本的8个杂交组合的平均杂交亲和指数为0.57%;以‘耶罗琳’为母本的6个杂交组合的杂交亲和指数均为0。所有杂交亲和的杂交组合中,其父本除LA百合‘正直’与川百合外,均为亚洲百合;亚洲百合与亚洲百合之间杂交有亲和性,而亚洲百合与东方百合、OT百合杂交均不亲和。‘小小的吻’部分杂交组合不亲和原因主要是花粉管未到达花柱底端所致;而以‘小小的吻’为母本的杂交亲和的组合中,花粉管均能到达子房,但其胚胎发育存在正常和异常2种情况。[结论] 百合杂交亲和性与父本花粉活力无关,主要与父母本亲缘关系有关,来自同一杂种系列的父母本杂交亲和性较高,亚洲百合‘小小的吻’最适合作为选育无花粉污染百合的母本。受精前生殖障碍是无花粉百合杂交不结实的主要原因。
Abstract:
[Objectives] This study aimed to study the relevant factors that affect pollen-free lily cross-compatibility,which aimed to lay the foundation for breeding new pollen-free lily cultivars.[Methods] Four lily cultivars(pollen-free ‘Little Kiss’ and ‘Easy Life’,anther dehiscence ‘Soft Waltz’,and ‘Yelloween’ with pollen agglomeration)were used as pollen receptor or female parent,11 lily germplasm resources were selected as pollen donor or male parent,and 33 hybrid combinations or crosses among them were carried out to investigate hybrid affinity. We used fluorescence microscopy and paraffin section to study the cytological reasons,which affect hybrid affinity in the crosses by using ‘Little Kiss’ as the female parent.[Results] In the male parents,pollen germination rate of oriental lily ‘Sorbonne’ was the highest(95.30%),and pollen germination rate of LA lily ‘Moselle’ was the lowest(6.79%). Among the 33 crosses,the average hybridization affinity index of ten crosses using ‘Little Kiss’ as the female was the highest,3.61%,and the hybridization affinity index of ‘Little Kiss’בTiny Padhye’ was the highest among all the crosses. The average hybridization affinity index of the nine crosses using ‘Easy Life’ as the female parent was 0.90%,and the hybridization affinity index of ‘Easy Life’בHonest’ was the highest. The average hybridization affinity index of the eight crosses using ‘Soft Waltz’ as the female parent was 0.57%. The hybridization affinity index of the six crosses using ‘Yelloween’ as the female parent was 0. The male parents of all the compatible crosses were Asian lily except LA lily ‘Honest’ and Lilium davidii,the crosses between Asian lily and Asian lily were compatible,and the crosses of Asian lily with oriental or OT lily were incompatible. The incompatibility of partial crosses of ‘Little Kiss’ was mainly caused by the fact that pollen tubes did not reach the bottom of the style,while pollen tubes could reach the ovary in the compatible crosses. There were two conditions as normal and abnormal status in the development of lily embryos.[Conclusions] Lily cross-compatibility was not related to pollen germination rate of the male parent,but mainly related to parental relationship. In addition,the hybrid affinity of the crosses with the same hybrids was high and the Asian lily ‘Little Kiss’ was the most suitable parent for breeding pollen-free lily. Pre-fertilization disorder was the main reason of sterility in hybridization breeding of pollen-free lily.
